Más de cincuenta años después de su descubrimiento inicial por Rita Levi Montalcini y Stanley Cohen y de la propuesta de la teorÃa neurotrófica, el factor de crecimiento nervioso (NGF) se ha convertido en el prototipo de una familia de moléculas biológicamente activas llamadas factores neurotróficos (NTF). Este libro aborda importantes avances en la investigación de NTF, desde la ciencia básica hasta la medicina clÃnica. Se centra principalmente en el NGF, pero también incluye capÃtulos individuales que tratan del factor neurotrófico derivado del cerebro (BDNF) y los ligandos de la familia del factor neurotrófico derivado de la lÃnea de células gliales (GDNF), que han atraÃdo un interés creciente en la comunidad de neurociencia debido a su diversos efectos en el cerebro normal y enfermo. En la primera parte del libro, los autores proporcionan los antecedentes necesarios para los siguientes capÃtulos y analizan los mecanismos y vÃas básicos de la transducción de señales de NGF. En las siguientes secciones, examinan la actividad regenerativa y la capacidad neuroprotectora del NGF durante el desarrollo y en tejidos normales y enfermos en la edad adulta y discuten el papel del NGF en la enfermedad de Alzheimer y la nocicepción. Además, se analiza más a fondo el papel del NGF en el procesamiento de la información sensorial y su influencia en el comportamiento. El libro concluye con una descripción general del potencial diagnóstico y terapéutico del NTF en los trastornos psiquiátricos y el tratamiento de la obesidad, asà como con un punto destacado de la investigación del NGF en medicina veterinaria. Muchos de los autores de este volumen participaron en la Segunda Reunión Internacional Rita Levi-Montalcini, celebrada en Bolonia, Italia, en 2019. More than fifty years after its initial discovery by Rita Levi Montalcini and Stanley Cohen and the proposal of the neurotrophic theory, nerve growth factor (NGF) has become the prototype of a family of biologically active molecules called neurotrophic factors (NTFs). This book addresses important advances in NTF research, from basic science to clinical medicine. It focuses mainly on NGF, but also includes individual chapters dealing with the brain-derived neurotrophic factor (BDNF) and ligands of the glial cell line-derived neurotrophic factor (GDNF) family, which have attracted increasing interest in the neuroscience community because of their diverse effects in the normal and diseased brain. In the first part of the book, the authors provide the necessary background for the following chapters and discuss the basic mechanisms and pathways of NGF signal transduction. In the following sections, they then examine the regenerative activity and neuroprotective capacity of NGF during development and in normal and diseased tissues in adulthood and discuss the role of NGF in Alzheimer's disease and nociception. In addition, the role of NGF in processing sensory information and its influence on behavior is further discussed. The book concludes with an overview of the diagnostic and therapeutic potential of NTF in psychiatric disorders and obesity management, as well as a highlight of NGF research in veterinary medicine. Many of the authors of this volume participated in the Second International Rita Levi-Montalcini Meeting, held in Bologna, Italy, in 2019. The book covers a wide range of important topics in past and current NTF research and will appeal to basic researchers and clinicians alike. |
